Several commonly used plant culture media

The success of tissue culture depends largely on the choice of medium. Different media have different characteristics, suitable for different plant species and inoculation materials. When carrying out tissue culture activities, various media should be understood and analyzed so that they can be selected for use. The following introduces several commonly used media for tissue culture.

1. MS medium. MS medium is a medium commonly used at present. It has a higher concentration of inorganic salts, which is very beneficial to ensure the mineral nutrition required for tissue growth and accelerate the growth of callus. Due to the high concentration of ions in the formula, even in the process of preparation, storage, and disinfection, even if some ingredients are slightly in and out, it will not affect the balance between ions. MS solid medium can be used to induce callus, or used to culture embryos, stem segments, shoot tips and anthers. Its liquid medium can be used for cell suspension culture to obtain obvious success. The amount and ratio of inorganic nutrients in this medium are suitable, which is sufficient to meet the nutritional and physiological needs of plant cells. Therefore, in general, no additional organic ingredients such as amino acids, casein hydrolysate, yeast extract and coconut milk are needed. Compared with the basic components of other media, the MS media has a high content of nitrate, potassium and ammonium, which is its obvious feature.

2. B5 medium. The main characteristic of B5 medium is that it contains lower ammonium, because ammonium may inhibit the growth of many cultures. After experiments, it was found that the callus and suspension cultures of some plants grew better on the MS medium than on the B5 medium, while other plants were more suitable on the B5 medium.

3. N6 medium. N6 medium is especially suitable for anther and pollen cultivation of cereals, and it is widely used at home and abroad. In tissue culture, White (While, 1963) medium and Nitsch (1951) medium are often used. They are similar in basic composition. White's medium is more suitable for tissue culture of woody plants due to the lower amount of inorganic salts.
